Curcumin, a naturally occurring polyphenolic compound isolated from Curcuma longa, exhibits significant anti-inflammatory, antioxidant, and therapeutic potential. Despite its promising pharmacological profile, the clinical application of curcumin is severely limited due to poor aqueous solubility, low gastrointestinal absorption, rapid metabolism, and swift systemic elimination, resulting in low bioavailability. Nanoparticle-based capsule formulations have emerged as an advanced drug delivery strategy to overcome these limitations. This review critically examines recent advances in curcumin nanoparticle capsule systems and their role in enhancing bioavailability and anti-inflammatory efficacy. Various nanocarriers, including polymeric nanoparticles, lipid-based systems, and polysaccharide-derived nanocapsules, are discussed with respect to formulation design, pharmacokinetic improvements, and mechanisms of action. Evidence from preclinical and emerging clinical studies demonstrates that nanoparticle encapsulation significantly improves curcumin stability, intestinal absorption, plasma concentration, and targeted delivery to inflamed tissues. Furthermore, nanocurcumin formulations exhibit superior suppression of pro-inflammatory mediators such as nuclear factor-κB (NF-κB), tumor necrosis factor-α (TNF-α), and interleukins when compared with conventional curcumin. Overall, curcumin nanoparticle capsules represent a promising approach for maximizing therapeutic efficacy and expanding the clinical applicability of curcumin in the management of inflammatory disorders.
