Abstract: Colocasia esculenta (L.), commonly known as taro, is a nutritionally and medicinally important plant containing diverse bioactive compounds. Among these, gallic acid is a key phenolic constituent known for its potent antioxidant, anti-inflammatory, and antimicrobial properties. The present study aimed to develop and validate a simple, precise, and reproducible High-Performance Thin-Layer Chromatography (HPTLC) method for the qualitative and quantitative estimation of gallic acid from the methanolic extract of C. esculenta. Chromatographic separation was achieved on silica gel 60 F₂₅₄ plates using a mobile phase of toluene: ethyl acetate: formic acid: methanol (3:3:0.8:0.2 v/v/v/v), and densitometric scanning was carried out at 254 nm. A distinct peak of gallic acid was observed at Rf = 0.51 ± 0.02, confirming its presence in the extract. The calibration curve showed good linearity in the range of 100–500 ng/spot, with the regression equation y = 1.41x − 44.333 and correlation coefficient R² = 0.998. The gallic acid content in the methanolic extract was found to be 357 ng/spot. Validation parameters, including recovery (98.3–99.6%), %RSD (<2.0), LOD (0.007 µg/spot), and LOQ (0.023 µg/spot), confirmed the method’s accuracy, precision, and sensitivity. Thus, the developed HPTLC method is reliable, cost-effective, and suitable for routine qualitative and quantitative analysis of gallic acid in Colocasia esculenta and related herbal formulations.
