Lung cancer, also known as lung carcinoma (since about 98–99% of all lung cancers are carcinomas), is a malignant lung tumor characterized by uncontrolled cell growth in tissues of the lungLymphomas and melanomas (from lymphoid and melanocyte cell lineages) can also rarely result in lung cancer. In 2005, the total number of new lung cancer cases in china was over 500,000. According to GLOBOCAN 2012,lung cancer is the most common malignancy and cause of cancer mortality in China, representing 21% of all cancers and 27% of all cancer-related deaths Lung cancer incidence and mortality is higher in eastern China and in urban areas, which has been attributed to westernization of lifestyle. EGFR is a transmembrane tyrosine kinase receptor, which is one of the ErbB family of receptors. The ErbB family includes EGFR (ErbB1), ERBB2 (HER2/neu), ERBB3 (HER3), and ERBB4 (HER4). Also known EGFR tyrosine kinase inhibitor, epidermal growth factor receptor inhibitor. EGFR kinase domain mutations including exon 19 deletion, L858R and T790M increase kinase activity of EGFR, leading to the hyperactivation of downstream signaling pathways including MAPK, PI3K/Akt/mTOR, and IL-6/JAK/STAT3 which promote tumorigenesis of NSCLC cells.
