Abstract: The present research was focused to develop the formulation containing combination of antidiabetic agent with natural antioxidant to manage diabetes-induced Alzheimer disease in elderly population with Type-2 diabetes. The thermoreversible in-situ gel formulations for nasal administration were developed to improve patient compliance, nasal bioavailability of drugs by increasing its nasal retention time and checked the effect of temperature on the preparation of formulation. The Metformin and Curcumin was used as model therapeutic agents. Combination of Curcumin with Metformin act synergistically on dyslipidaemia and oxidative stress, as well as increased PON 1 level which are major cause of AD. The in-situ gelation system was prepared by cold method with the use of Poloxamer P188, which has the property of thermo reversible gelation, Sodium alginate, Carbopol 940 and HPMC K100 were used as the mucoadhesive agents which will improve the absorption of drugs by increasing the contact time with nasal mucosa. Totally 12 formulations were prepared with Carbopol 940, Sodium alginate and HPMC K100 in varying concentrations. The prepared in-situ gel was evaluated for various quality control tests such as its gelation temperature, mucoadhesive potency, drug content, gelation time, viscosity and in vitro drug diffusion study.
