Abstract: Abstract Antimicrobials are produced by biofilm-forming marine isolate, Streptomyces sundarbansensis sp. nov., designated as MS1/7T in a novel polymethylmethacrylate enhanced surface area conico-cylindrical flask that provides significantly higher surface area and, also property that is favorable to surface attachment. Then, the process operating parameters are investigated using Response Surface Methodology wherein pH, temperature and rotational speed are considered as factors. Each of the three parameters are studied at three levels viz. high, medium and low –10, 9 and 8 for pH; 35°C, 30°C, 25°C for temperature and 160 rpm, 140 rpm and 120 rpm for rotational speed maintaining 100 mL total volume and 96h of incubation period. The peak antimicrobial activity (PAMA) determined are considered as a response. The optimum process conditions for antimicrobials production at 72h of incubation period are determined (pH 10, 30°C, 140 rpm). The model predicted the PAMA value of 41.7 mm at optimal value of process parameters (pH 10, 30.5°C, 140 rpm). The experiment conducted with the same optimum conditions that gave PAMA value i.e. 40.2 mm with 3.7 % error.
