The transdermal drug delivery system serves as a convenient means of administering drugs systemically through the skin barrier. However, the limited diffusion rate across the stratum corneum poses a significant challenge. To address this issue, the ethosomal delivery system has emerged as a recent innovation for drugs and herbal compounds. This approach offers advantages such as increased drug permeation, enhanced drug entrapment, and improved overall drug delivery. Ethosomes, characterized by their soft and malleable vesicular structure, consist mainly of phospholipids, ethanol, and water. The inclusion of ethanol enhances the fluidity of cell membrane lipids, facilitating greater penetrability of ethosomes through the skin. Once inside the skin, ethosomes permeate and fuse with cell membrane lipids, leading to effective drug release. Formulation methods involve both hot and cold techniques, with evaluation parameters encompassing size, shape, drug content, and zeta potential. Ethosomes have demonstrated successful outcomes in delivering various drugs, including Cyclosporine A, insulin, salbutamol, among others.
